The relationships between isoflavones and osteoporosis have been examined in many experimental studies of humans, animals, and cells in culture. Many studies have showed that isoflavones are related to reduce osteoporosis however there have few studies conducted the effect of isoflaveones on bone during growth period. This study was to investigate the ability of soy isoflavones, daidzein, genistein and mixture of two isoflavones to prevent bone loss in ovariectomized growing rats, compared to that of 17β -estradiol. Twenty one day old female Sprague-Dawley rats were accliminated for 3 days and then were sham operated or ovariectomized. They were divided into one sham, five ovariectomized (OVX) groups that were OVX, 10 ㎍/g b.w. of estrogen fed OVX, 100 ㎍/g body weight of genistein fed OVX, daidzein 100 ㎍/g b.w. fed OVX, and genistein and daidzein (100 ㎍/g b.w.) fed OVX. They were freely access to 76-G modified soy free normal diets and drinking water. Soy isoflavones and l7β-estradiol were given orally for 3 months. Upon days of 90, the femur bones were taken out to examine bone mineral density (BMD), bone strength, and calcium and phosphorous content. Serum was collected for analysis of Ca and phosphorous. The results showed that the body weight gain have no significantly difference among groups. The weight of organs, liver, spleen, kidney, and uterus in controls showed similar to that in experiment groups. The femoral volumetric BMD and BMC had not significant difference among groups but daidzein fed OVX group showed slightly increased BMD and BMC (p<0.06). Bone calcium content of rats was decreased by ovaiectomy. These findings suggest that isoflavones may prevent the bone loss in ovariectomized growing rats. Further research is needed to examine the magnitude, the responsible isoflavones and the mechanism of these effects.
