It has been reported that modifying effect of indole-3-carbinol (13C), one component of cruciferous vegetables, may be different at the time of carcinogen exposure and types of carcinogens. This study was investigated to examine the effect of pre-initiation treatment of 13C in N-methyl-N-nitrosourea (MNU)-induced rat mammary gland tumor model. Seventy-two female Sprague-Dawley rats were randomly divided into five groups. Animals of groups 1, 4 and 5 were given basal diet. Animals of group 2 were given the diet containing 100 ppm 13C and animals of groups 3 and 5 were given the diet containing 300 ppm 13C from 6 weeks of age. At 7 weeks of age, animals of groups 1, 2 and 3 were intraperitoneally injected with MNU, and those of groups 4 and 5 were treated with saline. All animals were killed at week 20 after carcinogen treatment. There were significant increases of tumor incidences in groups 2 and 3 compared to group 1 (P < 0.05). Tumor multiplicities of groups 2 and 3 were higher than those of group 1. The first occurrence of tumor was found in the 13C-feeding groups at week 8 after carcinogen treatment. Most of tumors were adenocarcinoma and there were no difference of tumor classification among the groups. Taken together, we conclud pre-initiation treatment 13C has promoting effect on MNU-induced mammary carcinogenesis and it suggests that it is important to set chemopreventive strategy of 13C in etiologic manner.
