An attempt was made to obtain a mutant strain from Nakayama strain of Japanese encephalitis virus cultivated in mouse brain by means of serial passage in embryonal cells of the snake (Agkistrodon halys Pallas). At first, the optimal temperature for the growth of the virus in embryonal cells of the snake was determined. Then, the titers of infectivity by mice LD, and plaque forming unit, and of hemagglutinin at each passage were measured. In addition to these assays, the immunogenicity of the virus was estimated at the 13th passage by cross-neutralization test between the snake-cell-grown virus and the mouse-grown virus and the rabbit sera immunized by them. The results can be summarized as follows: 1. An adequate temperature for the growth of Japanese encephalitis virus in embryonal cells of the snake was found to be 26C. 2. The mice Ldof the virus from the 1st to 5th passage in embryonal cells of the snake was relatively low but the titer remarkably increased from the 6th passage and thereafter. The plaque forming unit of the virus rapidly decreased from passage to passage until it reached almost nil at the 7th passage and shoved a slight recovery from the 12th passage. The plaques formed on the chick embryo fibroblast monolayer by the virus at the 13th passage in embryonal cells of the snake were shaped quite differently from those formed by the mouse-grown virus. The former were larger in size and irregular in shape with unclear margines. 3. The hemagglutinin titer of the virus grown in embryonal cells of the snake fell rapidly during the course of the serial passage to such a low degree that the titer was unable to be measured at the 6th passage. 4. The lowered plaque forming titer and the altered shape of the plaque at the 13th passage recovered their original level and shape when the virus was cultivated in Singhs mosquito cells after two successive subcultures. 5. According to the cross-neutralization test between the snake-cell-grown virus and the mouse-grown virus and the rabbit sera immunized by them, the immunogenicity of the virus did not seem to have changed at the 13th passage in embryonal cells of the snake.
