The purpose of the study was to investigate whether endothelial nitric oxide synthase(eNOS) gene polymorphisms are associated with changes in nitric oxide(NO) production with exercise training. Sixty healthy sedentary subjects(aged 50-70 yrs) completed 6 months of exercise training that consisted of endurance exercise at 70% of VO2max, 40 min/d, 3 d/wk. NO production was determined as the measure of plasma NOx(NO2- + NO3-) using a modified Griess assay. Genotype analyses for 27-bp repeat, G894T, and T-786C polymorphisms were performed using PCR/RFLP. At baseline, plasma NOx levels were not different among genotypes for 27-bp repeat, G894T, and T-786C eNOS gene polymorphisms. Systolic blood pressure(SBP) was significantly higher in 4b4b genotype, low-density lipoprotein cholesterol(LDL-C) was significantly higher in GG genotype, and % body fat was significantly higher in TC/CC genotype for 27-bp repeat, G894T, and T-786C polymorphism, respectively. After exercise training, there was a significant interaction between the T-786C polymorphism and changes in plasma NOx levels(p=0.007). The TT genotype group significantly reduced their plasma NOx levels from 20.7±0.9 µmol/L to 17.6±0.8 µmol/L(p=0.003), while plasma NOx levels did not change in the TC/CC genotype group. There was no genotype difference in the changes in NOx levels in 27-bp repeat and G894T polymorphism. The present results indicate that exercise-induced changes in NO production may be associated with the T-786C polymorphism.
