The ethionine resistance conferring gene (ERC1) was constitutively expressed under the control of the alcohol dehydrogenase gene promoter (ADC1p) and introduced into the chromosomes of an industrial polyploid strain of Saccharomyces cerevisiae by using the d-sequences of the Ty1 retrotransposon as the recombination site. d-Integrative cassette devoid of bacterial DNA sequences containing the ampicillin resistance gene was constructed that had the aureobasidin A resistance gene (AUR1-C) as the selection marker and ERC1 gene. The ERC1 gene was also employed as the selection marker in the d-integrative cassette lacking the AUR1-C gene. Industrial Saccharomyces cerevisiae transformed with these integrative cassettes exhibited strong resistance to DL-ethionine compared with nontransformants.
