Five fusion proteins between Z domains derivedfrom Staphylococcal Protein A and Green Fluorescent Proteinor Human Proinsulin were produced on the periplasm ofEscherichia coli. The efects of the molecular weight andamino acid composition of the translocated peptide, culturemedium composition, and growth phase of the bacterial culturewere analyzed regarding the expression and periplasmic secretionnot affected by the size of the translocated peptide (17-42 kDa) and that the highest periplasmic production valueswere obtained on the exponential phase of growth. Moreover,the highest periplasmic values were obtained in minimal medium,showing the relevance of the culture medium composition onsecretion. In silico prediction analysis sugested that withrespect to the five proteins used in this study, those that areprone to form αperiplasm.
