Background: Although murine models have been useful in understanding acute airway inflammation in asthma, the ovalbumin induced murine models of asthma have significant limitations in study of airway remodeling. Objective: We investigated the Dermatophagoides farinae (Der f) induced the asthmatic mouse model of airway remodeling. Method: Six-week-old female BALB/c mice were divided into four groups according to adjuvant. After BALB/c were sensitized to Der f and exposed to Der f via nasal inhalation a week for 6 weeks, repeated Der f exposure were done on 3 days/week for 6 weeks to induce chronic asthma. Twenty-four hour after the final antigen challenge, bronchial responsiveness was measured, and bronchoalveolar lavage and histologic examinations were carried out. Result: The number of inflammatory cells and eosinophil in bronchoalveolar lavage fluid (P<0.01), serum total IgE (P<0.01) and Der f-specific IgG1 (P<0.01) in all experimental group were significantly more increased than those in control group at 6 weeks and 12 weeks after immunization of Der f. Complete Freund's adjuvant group showed statistically significant differences in goblet cell numbers, thickness of peribronchial smooth muscle, degree of subeptithelial fibrosis (P<0.01) and airway hyperresponsiveness to methacholine (P<0.01) compared those with control group at 6 and 12 weeks. Conclusion: These data suggest that the Der f-induced asthmatic mouse model of airway remodeling using complete Freund's adjuvant may be useful in study of chronic asthma.
