Background: The use of growth factors in skin rejuvenationis emerging as a novel anti-aging treatment. While the role ofgrowth factors in wound healing is well established, their usein skin rejuvenation has only recently been to be studied andno controlled trials have been performed. Objective: Weevaluated the anti-aging effects of secretory factors ofendothelial precursor cells differentiated from human embryonicstem cells (hESC-EPC) in Asian skin. Methods: A totalof 25 women were included in this randomized, controlledsplit-face study. The right and left sides of each participant’sface were randomly allocated to hESC-EPC conditionedmedium (CM) or saline. To enhance epidermal penetration,a 0.25-mm microneedle roller was used. Five treatmentsessions were repeated at 2-week intervals. Results: Physician’sglobal assessment of pigmentation and wrinkles aftertreatment revealed statistically significant effects of microneedlingplus hESC-EPC CM compared to microneedlingalone (p<0.05). Skin measurements by Mexameter andVisiometer also revealed statistically significant effects ofmicroneedling plus hESC-EPC CM on both pigmentation andwrinkles (p<0.05). The only minimal adverse event wasmild desquamation in one participant. Conclusion: Secretoryfactors of hESC-EPC improve the signs of skin aging andcould be a potential option for skin rejuvenation.
