Biogenic polyamines have been shown previously to reduce oxidative stress in bacteria exposed to antibiotics. The polyamine protective effect could be due to either the scavenging of ROS or interfering with the binding of antibiotics to target macromolecules. To assess the contribution of each mechanism, a comparative study of the effect of polyamines (putrescine, cadaverine, spermidine, and 1,3-diaminopropane) and wellknown antioxidants (Trolox, thiourea) on the intensity of reactive oxygen species production in vitro as well as the survival of Escherichia coli cells exposed to antibiotics was carried out. Polyamines diminished the generation of superoxide in NADH/phenazine methosulfate reaction and neutralized H₂O₂ at least 20-fold less effectively than the antioxidants. Polyamines and antioxidants had no discernible effect on the antibacterial activity of β-lactam cefotaxime. Cadaverine reduced the decline in the CFU number caused by aminoglycoside amikacin by 2.7 orders of magnitude, while the other polyamines did by approximately 2 orders. The latter was 2 and 10 times higher than the effects of Trolox and thiourea, respectively. Under fluoroquinolone levofloxacin treatment, the protective effect of polyamines increased with an increase in their ability to bind DNA (cadaverine<putrescine<diaminopropane<spermidine). Addition of spermidine resulted in an increase in the CFU number by 2 orders, which was 10 times lower than the effect of Trolox and 10 times higher than that of thiourea. The protective effect of polyamines on bacteria exposed to antibiotics is rather related to their ability to bind macromolecules including those targeted by antibiotics than to their properties to scavenge reactive oxygen species.
