Plant cell culture system is one of the effective methods for producing target metabolites and recombinant proteins along with microbial system. Epidermal growth factor (EGF) is a protein that promotes the proliferation and growth of animal epithelial cells, and EGF production platform of these foreign proteins mainly uses the E. coli system. Recently, the importance of a plant-derived EGF protein production system is increasing due to the stability of recombinant proteins. In addition, in order to increase the production efficiency of the recombinant protein, studies are underway to transform the endoplasmic reticulum (ER) targeting amino acid sequence (KDEL), which targets the protein to the ER in plant cells, together with a gene of interest. Therefore, in this study, EGF and EGF-KDEL genes were introduced into the Centella asiatica callus in order to efficiently mass-produce EGF in the plant cell system. Two vectors were used for transformation in this study. The CaMV35S promoter, EGF gene, and NPTII gene were loaded in pKRE1, and the KDEL sequence was loaded in pKRE2 vector.
