In the present study, we report a novel conjugated phytochemical peptide compound synthesized to develop functional cosmetic ingredients. The synthesized peptide, hereafter referred to as fusion peptide, was produced by applying solid phase peptide synthesis procedure. The fusion peptide demonstrated promising skin whitening, antioxidant, wound healing, and hair regeneration stimulation properties. Cell viability was evaluated via the MTT assay utilizing human keratinocytes, human fibroblast, and human follicle dermal papilla cells. Analysis of the experimental data disclosed that fusion peptide exerted a significant impact on wound healing parameters, manifesting 63.27 activity when evaluated in the HaCaT cell line Furthermore, the fusion peptide exhibited potent DPPH(2,2-Diphenyl-1-Picrylhydrazyl) radical scavenging activity, reaching 72.23 compared to the negative control. In vitro assay, fusion peptide effectively inhibited tyrosinase activity and L-DOPA oxidation. The fusion peptide also suppressed mRNA expressions of the tyrosinase, TRP-1(Tyrosinase Related Protein-1), DCT(Dopachrome Tautomerase), and MITF(Microphthalmia-associated Transcription Factor) genes to 50.62, 56.95, 70.08 and 39.66 respectively, compared to untreated control. To assess the impact of fusion peptide on hair growth, we measured the mRNA levels of noggin, DKK-1(Dickkopf-related Protein-1), and SRD5A1(Steroid 5-Alpha Reductase-1). Following treatment with fusion peptide, there was a reduction in SRD5A1 and DKK-1 gene expression to 54.65 and 81.83 respectively, compared to the negative control. The fusion peptide treatment led to a significant upregulation of noggin gene expression, with a 123.58 increase compared to the negative control. Therefore, fusion peptide with its hair growth-promoting, antioxidant, and skin-whitening effects, is a promising candidate for a key ingredient in functional cosmetics.
