For identifying the cause of neuropathies such as Alzheimer''s disease, Parkinson''s disease and stroke, many research groups need a large amount of nerve cells. However it’s difficult to get the nerve cells from human, so many research groups use the SH-SY5Y neuroblastoma cells in vitro model. Nowadays, atomic force microscopy(AFM) is utilized in many studies for measuring the structural information and physical characteristics of soft material. So in this study, SH-SY5Y cells were chosen as the experimental specimens and a detailed surface potential by Kelvin Probe Force Microscopy(KPFM) and measurement of cell membrane hardness by Force spectroscopy before and after differentiating neuronal cell was carried out. And also plan to detect D-glutamic acid which is one of neurotransmitter emitted from differentiated SH-SY5Y cell by using AFM topography image. SH-SY5Y cells were differentiated by Retinoic acid(RA) treatment and differentiated SH-SY5Y cells are determined by western blot method. Cells were fixed by using 3.7% formaldehyde aqueous solution and then dried. KPFM, Force Spectroscopy and AFM are one of SPM system application. From KPFM images, we can conclude that when SH-SY5Y cells differentiate, their surface potential values are decreased. And hardness value of SH cells membrane after differentiation decreased about 25% than that without differentiation. In measurement of detecting neurotransmitter, we can detect D-glutamic acid by using antigen-antibody binding force. Thus in this study, SPM method gives us a useful guideline for SH cell differentiation. We expect that this method can be also extended for the application of differentiation from Mesenchymal stem cell(MSC) to nerve cell.
