This study aimed to provide practical data to develop functional cosmetics and hair growth agents by experimenting with the antioxidant activity of Dendropanax Morbifera and Rosa Multiflora root extracts and the growth factor expression effect of Human Follicle Dermal Papilla Cells (HFDPC). In terms of experimental methods, the study measured the DPPH scavenging activity by extracting active ingredients from the Dendropanax Morbifera and Rosa Multiflora roots using different extraction methods. As a result, the study conducted a cytotoxicity test on the HFDPC and identified the proliferation effect of the cells using the extracted natural ingredients by outstanding extract method, and researched the stimulation effect of growth factor protein expression through the western blotting procedure.
According to the research, the total polyphenol content was 101.925 mg/g in the Dendropanax Morbifera extract by Methanol-Sonication and 75.032 mg/g in the Rosa Multiflora root extract by supercritical extraction. The total flavonoid content in the Dendropanax Morbifera and the Rosa Multiflora root extracts was 26.110 mg/g and 14.365 mg/g, respectively, which was the highest.
According to the analysis, the DPPH scavenging activity of both Dendropanax Morbifera and Rosa Multiflora root extracts by Methanol-Sonication was 95.55% and 98.24%, respectively. The nitrite scavenging activity of the Dendropanax Morbifera extract by Methanol-Sonication was 31.21%, while that of the Rosa Multiflora root extract by supercritical extraction was 55.86%, at the highest. This was lower than the nitrite scavenging activity of the ascorbic acid (1 mg/mL) of 90~93%, which was used as a positive control group.
In terms of cytotoxicity of the Dendropanax Morbifera and Rosa Multiflora root extracts by Methanol-Sonication on the HFDPC using the MTT assay, no significant cytotoxicity was observed from both extracts within the range of treatment concentration of 10 ug/mL, 100 ug/mL and 500 ug/mL. The growth rate of cells was 92.1% for the Dendropanax Morbifera extract and 113.1% for the Rosa Multiflora root extract at the treatment concentration of 500 ug/mL.
The stimulation effect of growth factor expression against the control group (PBS) using the western blotting procedure was the 105% for the Dendropanax Morbifera extract and 126% for the Rosa multiflora root extract for PDGF-B; 110% for the Dendropanax Morbifera extract and 135% for the Rosa multiflora root extract for IGF; 94% for the Dendropanax Morbifera extract and 99% for the Rosa multiflora root extract for VEGF; 103% for the Dendropanax Morbifera extract and 100% for the Rosa multiflora root extract for KGF; 99% for the Dendropanax Morbifera extract and 106% for the Rosa multiflora root extract for hGH; and 98% for the Dendropanax Morbifera extract and 97% for the Rosa multiflora root extract for EGF.
This study found that Dendropanax Morbifera and Rosa multiflora root extracts had a high DPPH scavenging activity; no cytotoxicity on the HFDPC; a high proliferation effect of the HFDPC; and the stimulation effect of growth factor expression, verifying the feasibility of the extracts as a material for functional cosmetics and hair growth agents.
