Anti-dyslipidemic Effect of ACLL extract (Artemisiae Iwayomogii Herba, Curcumae Radix, Lycii Fructus, Ligustri Lucidi Fructus complex extract) on High Cholesterol Diet-induced Dyslipidemia in Mice and antioxidant in HepG2 cell

Objectives : This study was aimed to investigate the effect of ACLL extract (Artemisiae Iwayomogii Herba, Curcumae Radix, Lycii Fructus, Ligustri Lucidi Fructus complex extract) on oxidative damage in HepG2 cell and dyslipidemia in ApoE-knockout mice.
Methods : After treatment with ACLL extract in HepG2 cells, we measured polyphenol, flavonoids, DPPH radical, ABTS radical scavenging activity, ROS, GSH, GPx, GR, SOD, Catalase, free fatty acid, MDA, mRNA expression. ACLL extracts was administered at 200 mg/kg, and 400 mg/kg to Apoe mice, where hyperlipidrmia was induced by high cholesterol diet. After mice were treated with ACLL for 4 weeks, we measured AST, ALT, creatinine, BUN and lipid levels (total cholesterol, HDL cholesterol, LDL cholesterol, triglyceride), atherogenic index(AI) and cardiac risk factor(CRF), histologic change of liver and aorta.
Results :
1. ACLL didn''t show any cytotoxicity in HepG2 cells and any liver & renal toxicity in ApoE-knockout mice.
2. ACLL increased the DPPH and ABTS radical scavenging activity in HepG2 cells.
3. ACLL significantly decreased ROS, GPx and increased GSH, catalase activity compared to control group in HepG2 cells.
4. ACLL decreased free fatty acid, MDA, ACAT1, HMG-CoA reductase in HepG2 cells.
5. ACLL decreased total cholesterol, LDL-cholesterol, TG in serum.
6. ACLL decreased atherogenic index and cardiac risk factor compared to control group.
7. ACLL increased GSH, catalase, GPx in liver tissue compared to control group.
8. ACLL decreased free fatty acid, MDA, ACAT1, HMG-CoA reductase in liver tissue compared to control group.
9. As compared with the control group, fat infiltration in the liver tissue and aorta decreased in ACLL treated mice, and cell nucleus is located in central area.
Conclusions : We suggest that ACLL extract may have the control effects of dyslipidemia by improving antioxidant defense system and lipid metabolis
