Cheongnoemyeongsin-hwan (CNMSH), a herb medicine to treat cognitive
impairment, has been reported to have anti-inflammatory and antioxidant
efficacy in several experimental models. However, whether CNMSH can
ameliorate the amyloid-β (Aβ)-mediated cognitive impairments and
neurodegeneration in AD is unknown. To evaluate the protective effects of
CNMSH against Aβ-induced neuronal damage in vivo, CNMSH was
administered to mice for 2 weeks. The effects of dietary supplementation of
CNMSH on memory and learning skills in the Aβ-induced cognitive
impaired mice were explored using three different paradigms: the Morris
water maze, the T-maze spontaneous alternation task and the novel object
recognition task. Administration of CNMSH significantly attenuated Aβ
-mediated impairment of cognition and memory. CNMSH also effectively
attenuated Aβ-induced apoptosis and DNA damage, and inhibited the
expression of inducible nitric oxide synthase and cyclooxygenase-2. At the
same time, CNMSH reversed Aβ-induced effect on major AD markers (i.e.,
tau phosphorylation and β-secretase expression) and improved brain-derived
neurotrophic factor levels, suggesting that CNMSH improves Aβ-induced
memory deficits and cognitive impairment through regulation of these
proteins expression. Therefore, CNMSH has the potential to be a disease
modifying agent for the prevention of AD pathogenesis.
