Bone marrow stromal (BMS) cells are multi potent stem cells with the potential to differentiate into various cell types, including neuronal cells, mesodermal origin cells, bone, and cartilage. To investigate whether BMS cells can differentiate into neuronal cells, BMS cells were cultured in the presence of nerve growth factor (NGF), a major factor found in areas of spinal cord injury (SCI). NGF significantly increased the expression of NeuN, a representative marker of neuronal cells, on BMS cells. As an ill vivo model, severe SCI was induced in the rat by clip compression, and then the injured area was injected with BMS cells. BMS cell transplantation (2.5 × 10? cells/ea) produced a significant locomotor recovery effect in rats beginning four weeks after injury (Basso-Beattie-Bresnahan score 11.86 ± 4.6) compared with vehicle treatment (Basso-Beattie-Bresnahan score 6.8 ± 1.3) (p ＜ 0.05). The effect of recovery appeared six weeks after transplantation. It is postulated that transplanted BMS cells promote significant locomotor recovery in an injured spinal cord compared with recovery in control animals and that BMS cell transplantation has potential for the treatment of SCI.
