Caesalpinia sapan heartwoods against Dermatophagoidesfarinae and D. pteronysinus were assessed and comparedwith those evidenced by comercial benzyl benzoate andDET. The observed responses varied according to dosageand mite species. The LD50 values of the methanol extractsderived from C. sappan heartwoods were 6.13 and 5.44g/cm3 against D. farinae and D. pteronysinusFurthermore, the ethyl acetate fraction derived from themethanol extract was approximately 8.71 more toxic thanDEET against D. farinae, and 4.73 times more toxic againstD. pteronyssinus. The biologically active constituent fromthe ethyl acetate fraction of C. sappan heartwood extract waspurified via silica gel chromatography and high-performanceliquid chromatography. The structure of the acaricidalcomponent was analyzed by GC-MS, 1H-NMR, 13C-NMR,1H-13C COSY-NMR, and DEPT-NMR spectroscopy, andidentified as juglone (5-hydroxy-1,4-naphthoquinone). Basedon the LD50 values of juglone and its derivatives, the mosttoxic compound against D. farinae was juglone (0.076g/cm3), followed by benzyl benzoate (9.143g/cm3) and 2-methyl-1,4-naphthoquinone (40.0g/cm3). These results indicateC. sappan heartwoods is likelyto be the result of the effects of juglone. Additionally, juglonetreatment was shown to effect a change in the color of thecuticles of house dust mites, from colorless-transparent todark brownish-black. Acordingly, as a naturally occurringacaricidal agent, C. sappan heartwood-derived juglone shouldprove to be quite useful as a potential control agent, leadcompound, and house dust mite indicator.
