The purpose of this study was to investigate the anti-oxidant and anti-inflammatory effects of Daebunchungeum (DBCE) extracts on RAW 264.7 macrophage cells. Cell viability was measured by MTT assay at various concentrations of DBCE. The anti-oxidative effect of DBCE was investigated by measuring the 2,2-Diphenyl1-picrylhydrazyl (DPPH) and 2,2-azino-bis (3-ethylbenzothiazoline-6-sulfonic acid)diammonium salt (ABTS) scavenging activity. The amount of nitric oxide (NO) formed was measured. To investigate anti-inflammatory effects, the expressions of NF-κB, iNOS, COX-2 and TNF-α were evaluated via real-time PCR and western blotting by LPSinduced RAW 264.7 macrophages. Results showed that the cell survival rate of DBCE did not decrease significantly compared to the control group. In RAW 264.7 macrophages stimulated with LPS, NO production was significantly inhibited compared to the LPS treated group. While mRNA and protein expressions of iNOS, COX-2 and p-NF-κB were also significantly decreased compared to the LPS treated group. Besides, DBCE was found to have high DPPH and ABTS free radical scavenging ability. In conclusion, DBCE may be used as a new natural material for skin antiaging and anti-inflammatory agents.
