In the present research, the anti-oxidant and anti-inflammatory activities of rosemary oil in Normal Human Epidermal Keratinocytes (NHEKs) were investigated. The antioxidative effects of rosemary oil were measured using polyphenols, RT-PCR and western blot assay for HO-1 and NQO-1. The anti-inflammatory effects were measured by RT-PCR and western blot assay for COX-2, MMP-2, MMP-9 and iNOS. The rosemary oil showed high polyphenol content which is 0.5 mg/kg. Additionally, a very low cytotoxicity, i.e. 0.13% of the rosemary oil was observed in MTT assay. The rosemary oil also recovered cell viability in ROS accumulated NHEKs by UVB irradiation. Moreover, both transcription levels and protein levels of antioxidant enzyme, HO-1 and NQO-1 were increased by the rosemary oil in NHEKs. To note, UVB has inflammatory effects on NHEKs, which decreased the mRNA and the protein expressions of COX-2, MMP-2, MMP-9 and iNOS by treatment with the rosemary oil. These results suggest that due to the anti-oxidative and anti-inflammatory properties, rosemary oil has the potential to be used in functional cosmetics.
