Xerostomia, or dry mouth, is the most common long-standing problem in dental practice that is typically associated with impaired salivary gland function. Although many patients have been suffering from xerostomia, the etiology and effective treatment of xerostomia have not been fully demonstrated. Therefore, we established 3D salivary gland (SG) organoid modeling platform to investigate pathogenesis of xerostomia and discover therapeutic targets. SG organoid was generated from the mouse submandibular gland. With regulation of several factors, SG organoid grew and expanded stably with high capacity of proliferation and self-renewal. We found that ERK and Wnt/β-catenin pathway regulated proliferation and differentiation in SG organoid.
