Adoptive cell therapy became one of the most important cell-based immunotherapy. Natural killer (NK) cells have been considered as potential source for adoptive cell therapy among immune cells owing to the advantage of allogenic transfer. However, for clinical applications, strategies for NK cell expansion still should be developed due to limited ex vivo expansion and short half-life of NK cells. In this study, we designed 3D culture using alginate microcarrier as a scale-up strategy for enhancing cell proliferation and half-life for immune-therapy. Cell culture system through alginate scaffold leads to long-term expansion providing benefits for cell growth.
