Prunus yedoensis, known as Yoshino cherry, is common tree species in Korea. However, the fruit and its extract have not been examined for their effects on anti-aging despite its high antioxidant content including anthocyanin. Therefore, we investigated the pharmacological efficacy of P. yedoensis fruit 70% ethanol extracts (PYE) in extending lifespan and in inhibiting neurodegeneration by using Caenorhabditis elegans as a model animal. First, C. elegans were treated with various concentrations of the PYE: 3 μg/ml, 30 μg/ml, 300 μg/ml and 3 mg/ml. After the treatments, their lifespan, motility, and neurodegeneration were measured. In addition, expressions of genes involved in aging were measured and calculated by using quantitative PCR. As a result, the lifespans among all PYE-treated animals were increased. Animals treated with the highest concentration, 3 mg/ml PYE, showed a significant increase in lifespan compared to the control, with 29.77% increase. Generally, the motilities in the treated animals were improved. Moreover, the motility in 3 mg/ml PYE treated animals were improved by 30.18, 63.33, and 58.33% at day 5, 10 and 15 respectively. Accordingly, reductions of neurodegeneration in 300 μg/ ml and 3 mg/ml PYE treated animals were observed during aging. Quantitative PCR results also showed increase in expressions of genes involved in ILS pathway (daf-16), AMPK pathway (aak-2), mitochondrial quality control (ucp-4) and others (skn-1 and sod-2). These results that P. yedoensis fruit extend C. elegans lifespan with maintained motility and reduced neurodegeneraion in aging suggest a possibility of pharmacological use of P. yedoensis fruit for anti-aging purpose.
