Objectives
The aim of this study is to investigate the effects of Gaejigayonggolmoryo-Tang (GYT) on the ulcerative colitis induced by dextran sulfate sodium (DSS) in mice.
Methods
Colitis induced by free drinking of 5% DSS was tested on six-week-old male ICR mice. The mice were divided into two groups: prevention group and treatment group. Prevention group was once again divided into three groups -Sample group, Control group and Normal group. The ''Sample group'' was treated with GYT for 14 days before it was given 5% DSS for 5 days. The ''Control group'' was given water instead of the medication for 14 days before the 5 days of 5% DSS. The ''Normal group'' was not given 5% DSS for comparison purpose.
The treatment group was also divided into three groups: Sample group, Control group and Normal group. The ''Sample group'' was treated with GYT for 3 days after it was given 5% DSS for 5 days. The ''Control group'' was given water instead of the medication for 3 days after the 5 days of 5% DSS. The ''Normal group'' was not given 5% DSS for comparison purpose.
Result
Inhibiting effects on inflammatory enzymes investigated by cellular experiment show GYT inhibits the expression of COX-2 and iNOS, and the production of NO. Based on the primary cellular experiment, the effects on ulcerative colitis induced by DSS of mice tissues were investigated. The inhibiting effects on the damages of tissues, the expression of inflammatory enzymes such as NF-κB p65, COX-2 and iNOS, and apoptosis were confirmed. In cellular experiment, GYT was more effective in inhibiting the expression of COX-2 than the expression of iNOS. However, GYT was evidently effective on tissues to inhibit the expression of COX-2.
Conclusions
It is supposed that Gaejigayonggolmoryo-Tang has both preventive and therapeutic effects on ulceratice colitis induced by DSS. Also, it is worthy of research and development as a COX-2 inhibitor and a potential drug for inflammatory bowel diseases from natural products. Further investigations for exact mechanisms will be needed.
