This study researched the possibility of using low molecular weight peptides produced from soy sprout protein as raw materials for functional cosmetics by using protease produced from GRAS strain B. subtilis natto in order to produce peptides to have skin-whitening and anti-aging benefits. The protein content increased to produce functional peptides, which was 1.3 times higher than that of soy sprouts by-product protein from 40.79% to 53.20%. After optimizing fermentation conditions for the protease by isolating B. subtilis natto from cheonggukjang, the author of the study selected culture medium which contains soy sprout 3% extract, fructose 2.1% and casein 2.25%, showing 590.5 unit/mL of protease activation. As a fermentation condition, temperature of 35.8 ℃ and pH 8.2 were selected and the protease activity of 590.5 unit / mL was obtained and it was confirmed that prediction using the predicted value of 525.4 unit/mL and statistical optimization model was necessary. The protease activation of base/acid and enzymes, chemical degradation methods, showed that the proteins were degraded to peptides of less than 1.4 kDa in 6 N HCl, Alcalase, protamex and B. subtilis natto protease. The polyphenol contents of the physiologically active substance were 6 N HCl > commercial enzyme > B. subtilis natto protease > control. The control group was treated with distilled water and 0.2 M sodium phosphate buffer (PB). The contents of flavonoids increased, commercial enzyme > B. subtilis natto protease > 6 N HCl, and polyphenol and flavonoid contents were decomposed into low molecular weight peptides. In order to measure the whitening activation, tyrosinase inhibitory activity was measured, showing tyrosinase inhibitory activity, B. subtilis natto protease > commercial enzyme > 6 N HCl in that order. The collagenase inhibitory activity was measured in order to confirm anti-aging function and inhibitory activity showed B. subtilis natto protease > commercial enzyme > 6 N HCl > control in that order. It showed that the peptide of low molecular weight was effective for collagenase inhibitory activity as a result similar to tyrosinase inhibitory activity. Advantages to using B. subtilis natto protease can show its own superiority in many ways, including price competitiveness and safety, rather than using commercial enzymes when it comes to production of peptides which have low whitening and anti-aging function. In hence, B. subtilis natto protease can be used as a raw material for multi-functional cosmetics.
