An efficient and reproducible method for micropropagation of Gymnaster koraiensis was developed through direct shoot organogenesis. Explants obtained from active buds of its rhizomes were sterilized with three sterilization methods to avoid contamination. Cefotaxime at more than 250 ㎎ㆍL?¹ added to a culture medium perfectly removed explant contamination. Combinations of 0.05?0.4 ㎎ㆍL?¹ benzylaminopurine and 0.05?0.4 ㎎ㆍL?¹ α-naphthaleneacetic acid (NAA), and combinations of 0.05?0.4 thidiazuron (TDZ) and 0.0?0.5 ㎎ㆍL?¹ NAA were evaluated for multiple shoot induction. Three basal media (× 1/2 MS, MS, and × 2 MS) and three sucrose concentrations (30, 60, and 90 gㆍL?¹) were tested. MS basic medium and 30 gㆍL?¹ sucrose, respectively, was found to be the most suitable for inducing multiple shoots. MS medium containing a combination of 0.2 ㎎ㆍL?¹ TDZ and 0.1 ㎎ㆍL?¹ NAA was the best for multiple shoot proliferation after 9 weeks of in vitro culture (41 shoots per explant). Roots were successfully formed regardless of the presence of indole-3-butyric acid (IBA), though IBA at 0.1 ㎎ㆍL?¹ showed the best root development of G. koraiensis. They were transferred to three artificial growing media in a greenhouse and then acclimatized very well in soil mixture (peatmoss:perlite = 7:3, v/v).
