The effect of treadmill exercise on cell proliferation in the dentate gyros of the hippocampus was investigated via BrdU-specific immunohistochemistry. Male Sprague-Dawley rats were randomly divided into the three groups: the control group (n = 8), the low-intensity exercise group (n = 8), the moderate-intensity exercise group (n = 8). Animals of the exercise groups were put on running 5 days per week for 4 weeks. The regimen of the low-intensity exercise consisted with running at speed of 3 m/min for the first 5 min, 5 m/min for the next 5 min, then 8 m/min for the last 20 min, while the regimen for the moderate-intensity exercise was made with running at speed of 16 m/min for the first 5 min, 19 in/min for the next 5 min, then 22 m/min for the last 20 min. Single dose of 50 mg/kg 5-bromo-2`-deoxyuridine (BrdU) per day was injected intraperitoneally 1 hr before start of treadmill running during the commencement of the experiment. In the present results, weight gain of the rats in the moderate-intensity exercise group was significantly decreased compared to the control group and mild-intensity exercise group. The new cell formation was increased by the treadmill exercise. In the mild-intensity exercise group, BrdU-positive cells in the hippocampal dentate gyros was higher compared to the control group. In the moderate-exercise group, number of the BrdU-positive cells was increased compared to the control group and mild-intensity exercise group. In the present study, it can be suggested that treadmill exercise may facilitate recovery and regeneration from diseases of brain such as Alzheimer`s disease, Parkinson`s disease and injury such as that resulting from stroke by increase of new cell formation.
