A 2.5 kb aga gene encoding α-galactosidase (α-Gal) from Leuconostoc mesenteroides SY1 was cloned intopSJE, an E. coli-Leuconostoc shuttle vector. The recombinantplasmid, pSJEaga, was introduced into Leuconostoc citreumKCTC3526 (ATCC49370) by electroporation. Transcriptionlevel of aga was the highest in cels grown on rafinose (1%,glucose, and sucrose. Western blot using antibodies against α-Gal showed similar results to slot-blot results and enzymeactivity measurements. All the results indicated that the agawas successfully expressed in L. citreum and its transcriptionwas under the carbon catabolite repression (CR).
