Background：Colorectal cancer (CRC) is a clinically challenging malignant tumor in worldwide. As a natural product and a sesquiterpene lactone, costunolide (CTD), which has been reported to possess anticancer activities. However, the mechanism of regulation and precise target remain undiscovered in CRC. In the present study, we found that CTD inhibited CRC cell proliferation in vitro and in vivo by targeting AKT.
Methods: Anticancer effects of CTD were examined by MTT, soft agar, cell cycle and apoptosis analysis. Utilizing Phosphoprotein-specific antibody array and Costunolide-sepharose conjugated bead pull-down analysis for potential target confirm. Ubiquitination analysis was conducted for mechanism of CTD.
