PURPOSE: In this study, we investigated the effect of 1,2,3,4,6-penta-O-galloyl-β-D-glucose (PGG) on cell differentiation and dexamethasone-induced muscle atrophy in mouse C2C12 myoblast.
METHOD: C2C12 myoblasts were cultured with Dulbecco’s modified Eagle’s medium (DMEM), 10% fetal bovine serum (FBS) and were differentiated to myotube cells using a differentiation medium containing 2% horse serum for 4 days. A muscle atrophy model was generated by treating with 1 μM dexamathasone for 18 h. We determined the cell viability, microscopy, western blot, and quantitative real-time PCR analysis.
RESULTS: PGG had neither effect on cell viability of C2C12 myoblast at a concentration of 0-10 μM for 24 h nor protein and mRNA expression (Atrogin, Murf1, Myogenin, and MyHC).
CONCLUSIONS: These data indicate that PGG has no effect on cell differentiation and dexamethasoneinduced atrophy using C2C12 cells. Further studies are required to determine the interaction of nutrition and exercise on skeletal muscle atrophy.
