In this study, we investigatied the effect of regular physical activity(i.e., treadmil exercise) on the extent of apoptosis in mice skeletal muscles. female 11wk(n=36) were used. the mice(ICR) were housed two per cage in a temperature(20-23℃) and light controlled evironment with a 12:12h light-dark cycle and provided with food and water ad libitum. ICR(11wk) were trained(TR) 5days weekly for 4wk on treadmil. the exercise was carried out on a treadmill adapted in oder to permit 2 animals to run at the same. the average speed was of 24 and 25m/min, total endurance time was about 40min. the exercise group performed physical activity for 1h before being killed by decapitation. one section of quadriceps muscle was cut from the RㆍL low limbs, frozen in liquid nitrogen, and stored at -80℃ for further analysis. caspase-3, transcript contents were estimated by RT-PCR An DNA laddering was used to detect DNA fragmentation as an indicator of apoptosis, the results of the study can be summerized in the followings: non training group, caspase-3 mRNA was significantly increased on immediately of non training group when compared with rest in addition, it was substantially decreased in both immediately and recovery 5 hour when compared with that of rest, respectively. however, caspase-3 increased in recovery 5h of non training when compared with rest. the DNA laddering analyses indicated that the apoptotic index in quadriceps muscle of recovery 5h on pre training group. training group, caspase-3 was not different between non training group and training groups of rest in quadriceps muscle(p>0.05, data not shown). however, it was significantly decreased in both immediately and recovery 5 hour of training group when compared with non training group. DNA fragmentation in quadriceps muscle of the training group not found on all time impact.
