The purpose this study was to examine the effect of exercise training on plasma insulin concentration and GLUT-2 expression of pancreatic and liver tissues in pregnant rats. After confirming their pregnancy, the pregnant rats were randomly divided into four groups (n=10 in each group): control group, pregnancy group, pregnancy and low intensity treadmill exercise group, and pregnancy and moderate intensity treadmill exercise group. From the 15th day of pregnancy until delivery, pregnant rats in exercise groups were forced to run on treadmill for 30 min once a day. Plasma insulin level was measured, and GLUT-2 in pancreatic and liver tissue was measured using western blot analysis. The present result demonstrated that in pregnant rat, plasma insulin levels significantly increased compared to those of the normal rats, and GLUT-2 protein expression in the pancreatic and liver tissue was significantly decreased in pregnancy rats compared to non-pregnant rats. However, low and moderate intensity exercise significantly decreased plasma insulin levels and increased GLUT-2 expression in pregnant rats. In conclusion, the results of the present study suggest that exercise during gestational period may improve glycemic control by down-regulating plasma insulin level and up-regulating pancreatic and hepatic GLUT-2 in pregnant rats.
