The major house-dust mite allergen, Der f 2,stimulates the phospholipase D (PLD) in T lymphocytesfrom Dermatophagoides farinae specificallergic individuals. PLD activity increasedmore than two-fold in T cells from allergic patientscompared with those cells from normalcontrols with maximal responses within 30 m inafter exposure of Der f 2. A well-known PLDactivator PKC-α was found to be translocated tomembrane from cytosol in Der f 2-treated T cellsfrom Dermatophagoides farinae specific allergicindividuals. Down-regulation of PKC-α with phorbolmyristate acetate pretreatment for 24 habolished Der f 2-induced PLD activation. Ro320432, PKC inhibitor also reduced the effectsof Der f 2-induced PLD activation suggestingthat PKC-α acts as upstream activator of PLDin Der f 2-treated T cells. Taken together, thepresent data suggest that Der f 2 can stimulatePLD activity through the PKC-α activation in Tcells from Dermatophagoides farinae allergicindividuals
