A preparation (HIM-1) of herb mixture was designed to modulate the immune responses and to protect the hematopoietic and immune system against oxidative damage. The mixture of 3 edible medicinal herbs (Angelica gagantis Radix, etc.) was extracted with hot water and fractionated with ethanol. The preparation HIM-1 was made up with addition of ethanol-insoluble fraction yielded from a half of total water extract to the other half of total water extract. In vitro, the proliferation of murine lymphocytes and bone marrow cells by HIM-1 was due to its polysaccharide fraction. The lymphocytes and HL-60 cells were protected by HIM-1, its polysaccharide and ethanol fractions against oxidative stress induced by H2O2 or radiation. In mice challenged with SRBC, the formation of antibody secreting cells was enhanced by HIM-1. The GVH reaction was increased by HIM-1 in mice injected with allogeneic lymphocytes. HIM-1 administration prolonged the survival of mice inoculated with melanoma(B16F0) cells. In irradiated mice, HIM-1 administration increased the survival of bone marrow stem cells and promoted the repopulation of blood cells. In analysis of the regenerated lymphocyte subsets, B cells were firstly regenerated and then T cells were recovered in mice administrated with HIM-1. The antibody production against T-dependent antigen DNP-KLH was augmented by HIM-1 in irradiated mice. These results indicated that HIM-1, a preparation of the herb mixture, protected hematopoetic stem cells and promoted the immune responses against oxidative stress. Since the preparation of herb mixture is a relatively nontoxic natural product, it might be a useful modifier for prevention and control of diseases.
