This study examined whether the ethanol extract of lotus (Nelumbo nucifera) seeds (EL) has potential use functional food materials. Its antioxidant effect was evaluated by measuring its DPPH radical scavenging activity and reduction potential. In addition, its whitening activity was evaluated through its ability to inhibit tyrosinase, which binds to L-DOPA and contributes to melanin formation in the initial stage of melanogenesis. The EL treatment exhibited a dose-dependent increase in the total phenolic assay, DPPH radical scavenging, and reduction potential. The total phenolic content of EL at concentrations of 100, 250, 500, 750, and 1,000 μg/mL was measured as 4.32, 9.92, 18.42, 25.02, and 29.67 GAE μg/mL, respectively. The DPPH radical scavenging assay indicated inhibition of 4.28%, 9.56%, 11.02%, 24.77%, and 37.28% for the EL treatment at concentrations of 1, 5, 10, 25, and 50 μg/mL, respectively, compared to the control (no treatment). Furthermore, the reduction potential of EL was 0.79, 3.92, 8.11, 20.88, and 33.48 μM at concentrations of 1, 5, 10, 25, and 50 μg/mL, respectively. The whitening effect of the EL treatment showed inhibitory activities of 18.88%, 25.48%, 41.21%, 69.87%, and 89.65%. These results suggest EL could be used as a functional food material because of its antioxidant activity.
본 연구에서는 연자육 에탄올 추출물 EL을 제조하고 이의 총 폴리페놀 함량, DPPH 라디칼 소거 활성 그리고 환원력을 측정하였다. 또한, 멜라닌 형성 과정에서 초기 속도 조절에 관여하고, 멜라닌 생성에 있어 중요하게 작용하는 효소인 tyrosinase 저해 활성을 통해 미백 활성을 평가하였다. EL의 총 폴리페놀 함량은 농도가 증가함에 따라 증가하였으며 농도 의존적으로 DPPH 라디칼을 소거하였고, copper(Ⅱ) 이온에서 copper(Ⅰ) 이온으로의 환원력 또한 높은 수준을 나타냈다. 게다가 400 μg/mL 농도의 EL은 미백 활성 평가에서 미백 소재로 잘 알려진 arbutin과 유사한 높은 수준의 미백 활성을 나타내었다. 이러한 결과를 바탕으로 연자육의 강한 항산화 효과와 향후 추가적인 미백 활성 평가를 통해 이너뷰티와 바이오 기능성 식품의 소재로서의 가능성을 시사한다.
